Your best bet is to start with making sure that your keyword is at the beginning of your title. Try to include more tags that are related to your video and your keyword. Then write a nice juicy description in your video and naturally include variations of your keyword.

Once that is done you can start to build social links to your video from other websites, really helps if you have your own blog here. That should be enough to get you started. Of course comments, likes, dislikes, views, and rentation all play a big part but that's pretty much the basics that I would start with for now.
